England coach Andy Flower says the off-spinner has a badly bruised knee and could be out of action for two weeks, possibly returning for game six of the seven-match series in Sydney on February 2.
Swann received a blow to the left knee while batting in last Wednesday's first Twenty20 match against Australia in Adelaide.
"He has got a slightly strangely deformed kneecap on his left knee," Flower told reporters in Hobart today.
"He got hit and it is badly bruised and it is quite painful so he won't be playing in this game and might have to rest for 10 days to two weeks.
"That does give (James) Tredwell the opportunity depending on whether we want to pick two spinners or not."
Australia lead the series one-nil after their six-wicket win in Melbourne on Sunday night.
Broad broke down with a stomach-muscle strain during the second Ashes Test in Adelaide in December and is still doing recovery work.
Anderson was given time off after England's triumphant Ashes campaign, and missed the two T20 games and the first one-dayer. The right-arm swing bowler is expected to play in the fourth one-dayer against Australia in Adelaide on January 26.
"Stuart will be arriving out here on the 21st along with James Anderson," Flower said.
Flower said he didn't anticipate Broad to be fit to play in the one-day series.
But he added: "It is better for him to be out here rather than being in the middle of an English winter."
